Diafiltration (Buffer Exchange) Using Hollow Fiber Membranes instead of Dialysis Tubing - Automated Diafiltration

ثبت نشده
چکیده

Diafiltration is term used reflecting tangential flow filtration (TFF) method of “washing” or removing a permeable molecule (impurities, salts, solvents, small proteins, etc) from a solution in an accelerated method when compared to dialysis tubing. By selecting the appropriate membrane(s), it can also be used to exchange buffers, narrow particle size distributions, modify salt concentrations or maximize protein recovery in a clarification process. Buffer exchange, the process of using an ultrafiltration membrane to rapidly and gently replace one buffer of a protein solution with another, is one of the most common applications of diafiltration with Spectrum Laboratories, Inc. hollow fiber modules.

Diafiltration for Desalting or Buffer Exchange

Diafiltration is an ultrafiltration membrane technique for completely removing, replacing, or lowering the concentration of salts or solvents from solutions containing proteins, peptides, nucleic acids, and other biomolecules. The process selectively uses permeable (porous) membrane filters to separate the components of solutions and suspensions based on their molecular size. Smaller molecules ...

Evaluation of Novel Large Cut-Off Ultrafiltration Membranes for Adenovirus Serotype 5 (Ad5) Concentration

The purification of virus particles and viral vectors for vaccine and gene therapy applications is gaining increasing importance in order to deliver a fast, efficient, and reliable production process. Ultrafiltration (UF) is a widely employed unit operation in bioprocessing and its use is present in several steps of the downstream purification train of biopharmaceuticals. However, to date few s...

Diafiltration MALDI-TOF mass spectrometry method for culture-independent detection and identification of pathogens directly from urine specimens.

OBJECTIVES With the aim of rapid, culture-independent identification of microorganisms directly from urine specimens, we developed a diafiltration matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) method. METHODS In this procedure, urine specimens are desalted, fractionated, and concentrated prior to MS analysis. The analytic performance characteristi...

Fabrication and Modification of Thin-Film Composite Hollow Fiber NF Membranes

This study focuses on the preparation and modification of a thin-film composite (TFC) hollow fiber polyamide membrane fabricated by the interfacial polymerization of Piperazine (PIP) and trimesoyl chloride (TMC) on a porous polysulfone substrate. The effects of triethylenetetramine (TETA) and silica nanoparticles (SiO2) contents in the aqueous phase (as the additives) on the morphology and perf...

Detection and reduction of microaggregates in insulin preparations.

Insulin is an important biotherapeutic protein, and it is also a model protein used to study amyloid diseases, such as Alzheimer's and Parkinson's. The preparation of the protein can lead to small amounts of aggregate in the solution, which in turn may lead to irreproducible in vitro results. Using several pre-treatment methods, we have determined that pH cycling and diafiltration of the insuli...
